Psikiyatrist olarak araştırmalar yaptığı Cornell Üniversitesi'nde Otto Kernberg'in Cornell'e geldiği yıla kadar çalışmıştır. James F. Masterson, Otto Kernberg ile birlikte nesne ilişkileri teorisi iki Amerikalı öncüsü olarak kabul edilirler. Masterson'ın nesne ilişkileri teorisine önemli katkılar sağlayan kitapları çok sayıda dile tercüme edilmiştir. Gerçek kendilik alanında yazdığı ilk kitapları borderline ve narsisistik bozuklukların tedavisindeki önemli dönüm noktalarından biri olarak kabul edilmektedir.
