Mackesy, Northumberland'da doğdu ve büyüdü. Hexham'daki Radley College ve Queen Elizabeth Lisesi'ne gitti. Ayrıca iki kez kısa bir süre üniversiteye gitti, ancak her iki durumda da bir hafta içinde ayrıldı.
Mackesy kariyerine The Spectator'da karikatürist olarak başladı, ardından Oxford University Press'te kitap illüstratörü oldu. Ayrıca Comic Relief için müzayedeye çıkarılacak bir dizi çizim yaratmak için Love Really setinde Richard Curtis ile birlikte çalıştı; sevdiği hayır kurumuyla çalışmaya devam etti. Yaptığı çizimler üzerinde Mandela ile birlikte çalışan bir litografi projesi olan Nelson Mandela'nın Unity Series projesinde çalışmak üzere seçildi.
